He traveled to Ferrara at a young age to further his studies and secured his first position at the Accademia della Morte there. He later moved to Rome, where he served as organist at the Church of Santa Maria in Valicella and the Basilica of Santa Maria Maggiore. Queen Christina of Sweden, who was an important patron of musicians in Rome, was reportedly so impressed by Pasquinis exceptional playing skills that she praised him as the Prince of Music.
